AB OBJECTIVE: We report here a flow cytometry method dealing with the
quantitation of cells which die by apoptosis or necrosis, and its uses
in individuals at different stages of HIV infection. METHOD: The method
uses low concentrations of two DNA binding dyes tha allow to establish
selective regions for live, apoptotic and necrotic cells in a rat
thymocyte model. PBLC from patients and controls were analyzed either
freshly obtained or after activation with PWM. RESULTS: TABULAR DATA,
SEE ABSTRACT VOLUME. Results are expressed as %1SD of cells within
cytogram regions defining live apoptotic and necrotic cells; results of
PWM-stimulated cultures are in brakets. DISCUSSION: Individuals with HIV
infection show significant incrase of cell mortality atributable to
programmed cell death. Whereas a substantial proportion of these cells
display DNA fragmentation and DNA binding dye profiles characteristic of
apoptosis, a significant proportion exhibit features of DNA staining
similar to the observed when cells die by accidental death or necrosis.
